Overdose

Symptoms: possible diarrhea, weakness. In the event of an overdose or accidental ingestion of the drug by children, you should consult a doctor.

Treatment: symptomatic.

Contraindications

individual intolerance to the components of the drug or high sensitivity to anthraquinone drugs (for example, laxatives),

severe liver or kidney failure,

inflammatory bowel diseases,

lactose intolerance,

lactase deficiency,

glucose-galactose malabsorption,

pregnancy,

lactation period,

children's age (up to 18 years).

With caution: patients with irritation of the lower intestine - only in special cases.

Incompatibilities

Agdicerin is not recommended to be taken together with drugs or products that affect the consistency of the contents of the intestine (for example, a large amount of fiber).

Antacids reduce the absorption of the drug.

When taken simultaneously with antibiotics or chemotherapeutic drugs that affect the intestinal microflora, the frequency of adverse events from the intestine may increase.

Undesirable effects

With hypersensitivity to the components of the drug, allergic reactions of various degrees, rashes, and itching may occur.

From the digestive system: in the initial period of taking the drug, diarrhea, nausea, vomiting, and abdominal pain may temporarily occur. In this case, the dose should be reduced to 1 capsule per day.

There may be an intense staining of the urine, from yellow to brown (depending on the pH), which does not require a reduction in the dose (withdrawal) of the drug.

Pharmacodynamic properties

Diacerein inhibits the synthesis and activity of IL-1, which plays an important role in the development of inflammation, degradation and subsequent destruction of cartilage in osteoarthritis. Diacerein also slows down the formation of metalloproteinases (collagenases), which are involved in the process of damage to cartilage. With prolonged use, diacerein stimulates the synthesis of proteoglycans and does not affect the synthesis of PG.

Pharmacokinetic properties

After oral administration, diacerein is rapidly absorbed from the gastrointestinal tract and completely deacetylated to rein. Concomitant administration of the drug with food increases bioavailability by 25%. Cmax reina in the plasma is reached after 144 min. T1/2 of plasma is 255 min. The association of rein with albumin is almost 100%. With a single dose of 50 mg Cmax reaches 3.15 mg/l. With repeated administration of the drug Cmax increases due to the accumulation of the drug. Rein is excreted by the kidneys in unchanged form (20%) and in the form of glucuronide (60%) and sulfate (20%). Rein penetrates the placental barrier and the BBB.

Inside, after eating (whole, without chewing, with water).

The standard dosage is 1 capsule 2 times a day (morning and evening). The effect of the treatment should be expected in 2-4 weeks. Agdicerin is used continuously, for a long time, or in courses lasting at least 4 months. The duration of the break between courses of treatment is determined by the doctor.
